血紅素加氧酶-1 (HO-1)也稱為Hsp32,是血紅素加氧酶的誘導(dǎo)型異構(gòu)體,對(duì)血紅素的NADPH、氧和細(xì)胞色素P450還原酶依賴性氧化進(jìn)行催化,氧化為一氧化碳、亞鐵和膽綠素,膽綠素迅速還原為膽紅素。這些反應(yīng)產(chǎn)物具有重要的生理作用:一氧化碳是一種有效的血管擴(kuò)張劑,并被認(rèn)為是cGMP和血管緊張度的生理調(diào)節(jié)劑;膽綠素及其產(chǎn)物膽紅素是有效的抗氧化劑;“游離"鐵通過影響鐵調(diào)節(jié)蛋白(IRP)-1的構(gòu)象及其與mRNAs的5’-或3’-UTR中的鐵調(diào)節(jié)元件(IREs)的結(jié)合,增加氧化應(yīng)激并調(diào)節(jié)許多mRNAs(例如,DCT-1、鐵蛋白和轉(zhuǎn)移受體)的表達(dá)。三種已鑒定的血紅素加氧酶亞型是催化血紅素轉(zhuǎn)化為膽綠素和一氧化碳的血紅素加氧酶系統(tǒng)的一部分,分別是可誘導(dǎo)的HO-1或Hsp32,在腦和睪丸中大量存在的組成型HO-2,以及與HO-2相關(guān)但為不同基因產(chǎn)物的HO-3。
